For this, fatigue loaded as well as unloaded control samples were en-bloc stained in 1% basic fuchsin in ascending series of alcohols (80%, 90% and 100%) under 20-psi vacuum(Burr and Hooser, 1995). Further, bone samples were sectioned transversely into 200 µm thick sections using low speed diamond wafer saw and grounded to 100 µm thickness.

The blocks were stained as a whole in 1% basic fuchsin hydrochloride in a graded series of alcohols under vacuum according to a standard protocol (Burr and Hooser, 1995). After staining, the blocks were embedded using methlyl methacrylate.
