Localization of 5-HT4 receptor mRNA in rat brain by in situ hybridization histochemistry

Abstract

Oligonucleotide probes that recognize two cloned splice variants (5-HT4S and 5-HT4L) of 5-HT4 receptors were used to study by in situ hybridization the localization in rat brain of mRNA encoding these receptors. A probe common to both variants reveals high levels of transcripts in olfactory tubercle, some components of the basal ganglia (caudate putamen, ventral striatum), medial habenula and hippocampal formation. Similar patterns of distribution are obtained with probes that recognize each splice variant individually, suggesting that no dramatic differences exist in their respective regional distribution. Comparison of mRNA distribution with receptor distribution as visualized with [125I]SB 207710 indicates that 5-HT4 receptors are localized both somatodendritically in e.g. caudate putamen and on axon terminals in e.g. substantia nigra and globus pallidus.

    The data obtained in human hippocampal formation, where the 5-HT3 receptor is expressed prevalently by glutamatergic pyramidal neurons, are different from those obtained in rodents, in which this receptor is localized especially in inhibitory interneurons (Brady et al., 2007; Morales and Bloom, 1997). A high level of 5-HT4 receptor mRNA has been revealed in the rat hippocampal formation (Vilaro et al., 1996). Accordingly, immunohistochemical procedures have demonstrated that in the rat hippocampal formation the 5-HT4 receptor is located in the granule cell layer of the DG and pyramidal cell layer of the CA1 field (Egeland et al., 2011).

    The 5-HT4R couples to Gαs to positively link to adenylate cyclase in brain (for review, Bockaert, Claeysen, Compan, & Dumuis, 2004). Exhibiting a wide distribution throughout the CNS, the 5-HT4R is located in limbic areas (olfactory tubercle, PFC, hippocampus, amygdala), dorsal striatum, and NAc (Vilaró et al., 1996; Waeber, Sebben, Nieoullon, Bockaert, & Dumuis, 1994). The 5-HT4R localizes to terminals to facilitate DA, 5-HT, GABA, and acetylcholine release (for reviews, Bockaert et al., 2004, 2006).
