A high level of 5-HT4 receptor mRNA has been revealed in the rat hippocampal formation (Vilaro et al., 1996). Accordingly, immunohistochemical procedures have demonstrated that in the rat hippocampal formation the 5-HT4 receptor is located in the granule cell layer of the DG and pyramidal cell layer of the CA1 field (Egeland et al., 2011).

Exhibiting a wide distribution throughout the CNS, the 5-HT4R is located in limbic areas (olfactory tubercle, PFC, hippocampus, amygdala), dorsal striatum, and NAc (Vilaró et al., 1996; Waeber, Sebben, Nieoullon, Bockaert, & Dumuis, 1994). The 5-HT4R localizes to terminals to facilitate DA, 5-HT, GABA, and acetylcholine release (for reviews, Bockaert et al., 2004, 2006).
