However, distribution is also affected by other tracer properties such as shape, surface charge and number of Tc atoms per colloid particle, specific activity, depth and site of injection, as well as lymphatic drainage and availability of macrophages in the lymph node(s) [3]. Most European centres use Tc99m-nanocolloid (mean particle size 5–80 nm), whereas microfiltered Tc99m-sulphur colloid is usually used in the United States (mean particle size 100–220 nm) and Tc99m-labelled antimony trisulphide in Australia (mean particle size 3–30 nm) [4].

These factors include the particle size [4,5] and the surface properties of the nanocarriers [6,7] as well as the administration route [8,9]. For example, several studies have shown that, following subcutaneous administration, small nanocarriers (size close to 100 nm) can be efficiently drained from the interstice of the injection site into the lymphatics and be retained in the lymph nodes [10,11].
