Structure and functional expression of the cloned rat neurotensin receptor

Abstract

A functional cDNA clone for the rat neurotensin receptor was isolated by combining molecular cloning in an RNA expression vector with an elect rophysiological assay in Xenopus oocytes. The neurotensin receptor consists of 424 amino acids with seven putative transmembrane domains and belongs to the family of G protein-coupled receptors. The cloned receptor expressed in mammalian cells or in Xenopus oocytes shows a selective and high-affinity binding to neurotensin peptides and undergoes potent desensitization by repeated application of neurotensin. The neurotensin receptor mRNA is expressed in both the brain and the peripheral tissues at different levels. This investigation discloses the molecular nature of the neurotensin receptor, which mediates the diverse neuronal and peripheral actions of neurotensin by effecting the G protein-associated second messenger system.

      Subsets of VTA DA neurons have begun to be distinguished via their expression of neuropeptide receptors (Heymann et al., 2020; Perez-Bonilla et al., 2020; Woodworth et al., 2018), which hints at their potential contributions to energy balance, given that neuropeptides may be orexigenic or anorectic. One subset of VTA DA neurons co-expresses the Gαq protein-coupled neurotensin receptor-1 (NtsR1), which binds the neuropeptide Neurotensin (Nts) (Tanaka et al., 1990). Nts exerts diverse, site-dependent regulation of pain, sleep, locomotor activity and ingestive behavior, but within the VTA Nts suppresses feeding and promotes locomotor activity - dual behaviors that could support weight loss (Cador et al., 1986; Elliott and Nemeroff, 1986; Schroeder and Leinninger, 2018; Torruella-Suárez and McElligott, 2020).
