Methods: CITED1-siRNA and negative control were successfully transfected into thyroid cancer K1 cell line. Real-time PCR and Western blot were used to detect the knock down efficiency. CCK-8 method was used to detect cell proliferation, FACS method was used to detect cell cycle, and wound healing assay was carried out, cell apoptosis was analysed by flow cytometry. Results: Slow virus successfully infected K1 cells, mRNA and protein detection showed that the CITED1 gene was knocked down successfully. After CITED1 gene knocked down, the cell proliferation rate significantly inhibited, G0/1 phase cells increased, G2/M and S phase decreased, cell apoptosis rate was significantly higher, and the relative migration abilities of CITED1-siRNA group at 12, 24h after transfection were significantly lower than those in normal control group and negative control group. Conclusions: CITED1 gene may affect the occurrence and development of thyroid papillary cancer by promoting tumor cell proliferation and migration,and inhibiting apoptosis.
