We have evaluated the glutamine addiction in breast cells, MCF-7, by PET imaging with 18F-(2S,4R)4-fluoroglutamine in breast tumor cell, MCF-7, which are known to have c-Myc gene over expression. Methods: 18F-(2S,4R)4-fluoroglutamine was synthesized using the two-step chemical reaction and the radiochemical purity was determined by radio-high-performance liquid chromatography (Radio-HPLC). The stability of this tracer in PBS and saline buffers was measured. The Myc gene expression in MCF-7 cells was measured using qPCR. The correlation between Myc gene levels and in vitro cell uptakes of 18F-(2S,4R)4-fluoroglutamine in MCF-7 cells was compared with that of 18F-fluorodeoxyglucose (18F-FDG). In vitro biodistribution and micro-PET imaging studies of 18F-(2S,4R)4-fluoroglutamine were performed in MCF-7 bearing BALB/c nude mice. Results: 18F-(2S,4R)4-fluoroglutamine was synthesized with high radiochemical purity (&gt;95 %) and the tracer was stable in PBS and saline buffers during 2 hr incubation. Myc gene was up regulated in MCF-7 cells as measured by qPCR. The cell uptake of 18F-(2S,4R)4-fluoroglutamine was higher in MCF-7 cells when compared to that of 18F-FDG. Both of the biodistribution and micro-PET images showed higher tumor accumulation of 18F-(2S,4R)4-fluoroglutamine in MCF-7 tumor models. Conclusion: 18F-(2S,4R)4-fluoroglutamine may be useful as a metabolic radiotracer for noninvasive imaging of breast cancer, in which the Myc gene expression is up regulated.
