The objective of this study was to monitor the effectiveness of neural transplantation therapy in Parkinson's Disease rat model using 4-[18F]-ADAM and microPET. Methods 4-[18F]-ADAM was synthesized as previously reported (Peng et al., 2008). The PD rat model (n=18) was induced in male Sprague-Dawley rats by unilateral injection of 6-hydroxydopamine (6-OHDA , 5μg/μl in PBS) into the right median forebrain bundle. Motor function was evaluated 14 day after 6-OHDA lesion followed by amphetamine (5 mg/kg, i.p.) injection. The rotational score &gt;300/hr was considered to be PD. Three weeks after 6-OHDA lesion, six PD rats were injected with 4-[18F]-ADAM and imaged with a micro-PET R4 scanner. Images were collected from 60-90 min post injection and ROIs were drawn on both sides of midbrain, thalamus, hypothalamus, striatum and frontal cortex. Three weeks after 6-OHDA lesion, another six PD rats were sacrificed for immunohistochemistry (IHC) studies. The IHC images were quantified with Image-Pro Plus v. 6.0, and the ratios of ROI to control region were determined. The other six PD rats were ipsilaterally transplanted with E14 embryonic mesencephalic slurry (~5×105 cells) in striatum and imaged with PET four weeks after transplantation. The specific uptake ratio (SUR) of 4-[18F]-ADAM were expressed as ROI-cerebellum/cerebellum. Results Three weeks after 6-OHDA injection, the decrement of 4-[18F]-ADAM uptake in the lesioned striatum was 32% (by PET) and 45% (by IHC), respectively, compared to the unlesioned striatum. Four weeks after transplantation, 4-[18F]-ADAM uptake in the lesioned striatum was recoverd by 20%. The rotational score was 450 and 70/hr, respectively, before and after graft. Conclusions SERTs are involved in PD, and 4-[18F]-ADAM-PET may be an useful tool for monitoring the effectiveness of neural transplantation therapy in PD.
