rhBMP-2m up-regulated c-Myb gene expression in bone marrow cell of irradiated mice

Objectives: Objectives: Bone Morphogenetic proteins (BMPs), members of transforming growth factor-β (TGF-β) super family are a group of related protein capable of inducing stroma cells to differentiate into the formation of cartilage and bone, BMP plays an important role in the development of hematopoietic system also: if there isn’t BMP , the blood island formation was delayed or inhibited in the stage of embryo of xenopus leaves. Methods: BMPs now regarded as multifunctional cytokines. rhBMP-2m can be used to treat the injury of acute irradiation sickness in mice. We got rhBMP with gene technique, and found there were new hematopoietic points at the place where BMP induced bone and cartilage formation. We observed the effect of BMPs on the hematopoietic injury of acute radiation sickness in adult mice, and use RT-PCR and RNA dot blot hybridization technique. In bone marrow cells of rhBMP-2m group c-Myb oncogene expression was higher than that expression in irradiation and normal goups in RNA dot blot hybridization experiment. Results: We found the expression of c-Myb oncogene in the bone marrow cells of normal contral, irradiation control and rhBMP-2m treatment group on day 20 after a whole-body irradiated with 7.5Gy 60Co-γ ray, there was no evident difference among the 3 groups. But on day 10, the c-Myb oncogene of rhBMP-2m treatment group up-regulated, after rhBMP-2m was injected into murine, the expression was higher than that in irradiation and normal groups. Conclusions: The c-Myb oncogene expression is up-regulated in bone marrow cell of irradiated mice. Assisted by rhBMP-2m, the c-Myb oncogene promoted the recovery of hematopoietic function after irradiation.
