ABSTRACT
The uptake, incorporation and release of 14C-labeled amino acids and 51Cr were studied in Ehrlich ascites tumor cells. While the rates of uptake of radioactive materials into the cells and lysed cell supernatant were not significantly different, 14C-amino acids were recovered in greater quantity from the lysed cell centrifugate and the TCA-precipitable material. The nondialysable 14C-labeled material also had higher activity than that labeled with 51Cr. The 51Cr- and 14C-labeled materials were released from cells over a range of osmolalities in a pattern parallel to that of cell death as measured by trypan blue exclusion. Cellular exudation of 51Cr-labeled material and 14C-amino acid-labeled material, associated with cell adhesion, were similar and increased with time. The results confirm the usefulness of 51Cr labeling as an accurate marker of cytolysis and cell exudation.
